چکیده

Variation at regulatory elements, identified through hypersensitivity to digestion by DNase I, is believed to contribute to variation in complex traits, but the extent and consequences of this variation are poorly characterized. Analysis of terminally differentiated erythroblasts in eight inbred strains of mice identified reproducible variation at approximately 6% of DNase I hypersensitive sites (DHS). Only 30% of such variable DHS contain a sequence variant predictive of site variation. Nevertheless, sequence variants within variable DHS are more likely to be associated with complex traits than those in non-variant DHS, and variants associated with complex traits preferentially occur in variable DHS. Changes at a small proportion (less than 10%) of variable DHS are associated with changes in nearby transcriptional activity. Our results show that whilst DNA sequence variation is not the major determinant of variation in open chromatin, where such variants exist they are likely to be causal for complex traits.

P-46: Effects of Different Doses of Ethanol on Sperm Parameters, ChromatinStructure and Apoptosis in Adult Mice

Background: Chronic ethanol abuse causes reproductive organ failure and infertility in both humans and laboratory animals. Since sperm has a critical role in reproductive function, the objective of this unique study was to evaluate the effects of different doses of ethanol on sperm parameters, chromatin structure and apoptosis in adult mice. Materials and Methods: A total of 36 adult male mice ...

Examining the Causes and Consequences of Context-Specific Differential DNA Methylation in Maize.

DNA methylation is a stable modification of chromatin that can contribute to epigenetic variation through the regulation of genes or transposons. Profiling of DNA methylation in five maize (Zea mays) inbred lines found that while DNA methylation levels for more than 99% of the analyzed genomic regions are similar, there are still 5,000 to 20,000 context-specific differentially methylated region...

P-75: The Protective Effect of Vitamin C on Sperm Parameters and Chromatin Condensation in Diabetic Mice

Background: Diabetes mellitus (DM), primary or idiopathic, is a chronic disorder of the carbohydrate, lipid and protein metabolism, characterized through hyperglycemia and glycosuria. DM can involve male reproductive function at many levels. Vitamin C ( ascorbic acid) is one of necessary elements in human and mammalian diet that is very important in fertility. Vitamin C prevent from gamets dama...

Variation in DNA-Damage Responses to an Inhalational Carcinogen (1,3-Butadiene) in Relation to Strain-Specific Differences in Chromatin Accessibility and Gene Transcription Profiles in C57BL/6J and CAST/EiJ Mice.

BACKGROUND The damaging effects of exposure to environmental toxicants differentially affect genetically distinct individuals, but the mechanisms contributing to these differences are poorly understood. Genetic variation affects the establishment of the gene regulatory landscape and thus gene expression, and we hypothesized that this contributes to the observed heterogeneity in individual respo...

P-12: The Improvement of Sperm Parameters and Chromatin Quality by Vitamin E Supplement in Mice

Background: Vitamin E is believed to be the primary component of the antioxidant system in spermatozoa. There is a relationship between activity of these antioxidant and function of sperm. Vitamins E and C which are belong to non-enzymatic antioxidant are used as a supplemented drug to improve sperm quality and male fertility. The present study was carried out to investigate the positive effect...
